Autism and cancer evidence: autism baseline unchanged; lung-cancer survival signal persists

No new replicated causal autism result or official surveillance report established a single cause, and high-quality evidence still does not support vaccines as an autism cause. BioNTech reported updated Stage 1 overall-survival benefit for investigational gotistobart in the randomized Phase 3 PRESERVE-003 trial for squamous non-small-cell lung cancer that progressed after immunotherapy and chemotherapy. The pivotal Stage 2 remains ongoing; the drug is not FDA-approved and is not a cure.
Central finding
- Earlier randomized Stage 1 data reported HR 0.46 with a 95% CI of 0.25–0.84.
- Gotistobart is investigational, Stage 2 is ongoing and no FDA approval exists.
Evidence and limits
Previously published Stage 1 data at 14.5 months' median follow-up showed a death hazard ratio of 0.46 (95% CI 0.25–0.84), with median survival not reached versus about 10 months for docetaxel. The September 14 account says benefit persisted but does not provide complete updated medians, absolute survival, adverse-event rates or independently replicated data. Company and conference reporting is not regulatory approval.
- Complete updated absolute effects and toxicity were not available in the new report.
- No single autism cause has been established.
Why it matters
The signal could matter for a difficult post-immunotherapy population, but treatment decisions require the complete pivotal result, toxicity, absolute benefit, peer review and FDA assessment.
What changed
BioNTech presented an updated Stage 1 survival signal; no autism-causation change occurred.
What comes next
PRESERVE-003 Stage 2 enrollment and results; complete medians, confidence intervals, absolute survival, adverse events, peer review, independent replication and FDA action; replicated autism research.
U.S. strategic assessment
High interestPatients and families are harmed when preliminary associations or limited treatment results are promoted as established causes or cures.
Objectives
- Protect U.S. security, economic resilience, and citizens using measurable, lawful action.
Strategy
- Use primary records, allied coordination, market evidence, and independent verification to distinguish action from rhetoric.
Outcomes
- No result crossed the publication threshold during the window.
Risks
- Evidence searches are constrained by publication timing and access.
